      目的检测正常妊娠早期人工流产和复发性自然流产(recurrent spontaneous abortion,RSA)患者蜕膜巨噬细胞中免疫球蛋白样凝集素(sialic acid-binding immunoglobulin-like lectins,Siglec)-11的表达差异,探讨母胎界面巨噬细胞中Siglec-11的变化对母胎免疫耐受微环境平衡的作用。方法84例正常早孕人工流产时(对照组)、84例RSA患者清宫时(RSA组)取其蜕膜;提取核酸和蛋白质,实时定量荧光PCR法分析RSA组和对照组Siglec-11 mRNA的表达水平;免疫组化和免疫荧光检测Siglec-11分子在RSA患者蜕膜巨噬细胞上的原位表达情况及其与对照组的差异,通过高通量组织芯片的方法验证;ELISA法检测RSA患者和正常对照组血清中辅助型T细胞1(T helper 1 cell,Th1)/Th2细胞因子的表达水平变化。结果与对照组比较,Siglec-11在RSA组mRNA和蛋白水平上显著降低(P<0.05);且RSA组Th1型细胞因子表达升高,Th2型细胞因子表达降低(P<0.05)。结论Siglec-11的表达降低影响巨噬细胞功能,可能与母胎免疫平衡被破坏及RSA的发病有关。

      ObjectiveTo determine the expression and role of sialic acid-binding immunoglobulin-like lectins (Siglec)-11 in decidua macrophages in recurrent spontaneous abortion (RSA). MethodsThe decidua were collected from 84 normal pregnant women undergoing induced abortion (control group) and 84 RSA women undergoing early abortion (RSA group). Total RNA were extracted from decidua tissue. The expression of Siglec-11 in decidua macrophages at mRNA was analyzed by real-time quantitative PCR. Immunohistochemistry and immunofluorescence were used to detect the in situ protein expression of Siglec-11 in the decidual macrophages of RSA patients and its difference with the control group. A high-throughput tissue chip was constructed for verification. The serum expression of T helper 1 cell (Th1) and Th2 cytokine was measured by ELISA. ResultsThe mRNA and protein levels of Siglec-11 in decidua macrophages in RSA group is significantly lower than those in control group (P<0.05). The expression of Th1 cytokine in RSA group is significantly higher than those in control group and the expression of Th2 cytokine in RSA group is significantly lower than those in control group (P<0.05). ConclusionDecreased expression of Siglec-11 affects macrophage function, which may be related to the destruction of maternal fetal immune balance and the onset of RSA.
