      目的探讨多西紫杉醇诱导卵巢癌细胞凋亡及对磷脂酰肌醇 3-激酶(phosphoinositide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)信号通路的影响。方法选择人卵巢癌细胞株HO-8910,按照多西紫杉醇药物浓度分为Ho组(0 μg/mL)、Hd组(10 μg/mL)、Hz组(20 μg/mL)、Hg组(80 μg/mL),RT-PCR、Western Blot法分别检测PI3K、AKT水平、蛋白表达变化,Transwell法、MTT法检测HO-8910细胞的迁移、活力变化,流式细胞仪测定HO-8910细胞凋亡率。结果Ho组PI3K、AKT水平高于其他3组,4组差异有统计学意义(P<0.05);Ho组PI3K、AKT蛋白最高,其余3组PI3K、AKT蛋白表达从低至高依次为Hg组、Hz组、Hd组,4组差异有统计学意义(P<0.05);Hg组细胞凋亡率最高,其次为Hd组、Hz组、Ho组,4组调亡率差异有统计学意义(P<0.05);Ho组细胞迁移能力最高,其次为Hd组、Hz组、Hg组,4组细胞迁移能力差异有统计学意义(P<0.05);Ho组细胞增殖能力最高,其次为Hd组、Hz组、Hg组,4组增殖能力差异有统计学意义(P<0.05)。结论多西紫杉醇可促进HO-8910细胞大量凋亡,其作用机制可能为通过抑制PI3K/AKT信号通路活性来实现的。

      ObjectiveTo investigate the effect of docetaxel on apoptosis of ovarian cancer cells and phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT) signaling pathway. MethodsThe human ovarian cancer cell line HO-8910 was selected and divided into Ho group (0 μg/mL), Hd group (10 μg/mL), Hz group (20 μg/mL), Hg group (80 μg/mL) according to concentration of docetaxel. RT-PCR and Western Blot methods were used to detect PI3K, AKT levels and protein expression changes. Transwell and MTT methods were used to measure the migration and proliferation viability of HO-8910 cells. Flow cytometry was used to determine the apoptosis rate of HO-8910 cells. ResultsThe levels of PI3K and AKT in the Ho group were higher than those in the other three groups, and the difference among the four groups was significant (P<0.05). The Ho group had the highest PI3K and AKT protein levels, and the remaining three groups had the highest levels of PI3K and AKT protein expression from low to high in the Hg group, Hz group, and Hd group, and the difference among the four groups was significant (P<0.05). The Hg group had the highest apoptosis rate, followed by the Hd group and the Hz group, and the Ho group had the lowest apoptosis rate. The difference of apoptosis rate among the four groups was significant (P<0.05). Ho group had the highest cell migration ability, the remaining three groups had lower cell migration ability, Hg group had the lowest migration ability, Hz group had lower migration ability than Hd group, Hg group had lower migration ability than Hd group, and there was a significant difference among the four groups (P<0.05). Followed by the Hd group and the Hz group, the Hg group had the lowest cell proliferation ability, followed by the Hd group, Hz group, Hg group, the Hz group had a lower value than the Hd group, and the difference among the four groups was significant (P<0.05). ConclusionDocetaxel can promote a large number of apoptosis in HO-8910 cells, and its mechanism may be achieved by inhibiting the activity of PI3K/AKT signaling pathway.
