      目的 研究痔断根片抗炎、镇痛、通便和止血作用,对结肠黏膜组织水通道蛋白1(aquaporin1,AQP1)表达的影响,明确其药效特点,为临床定位提供可靠的实验依据。方法 采用二甲苯诱导的耳肿胀急性炎症模型研究其抗炎作用;采用乙酸诱导的疼痛模型研究其镇痛作用;采用洛派丁胺诱导的便秘小鼠研究其对肠胃蠕动的影响;采用实时荧光定量PCR(qRT-PCR)检测对各组便秘小鼠结肠黏膜中AQP1mRNA表达的影响;采用小鼠止血、凝血试验研究其止血作用。结果 痔断根片可以减轻二甲苯引起的小鼠耳廓肿胀,且肿胀抑制率随剂量增加而提高;能减少乙酸引起小鼠疼痛的扭体次数;缩减便秘小鼠首粒黑便的排出时间,增加5 h内黑便粒数和粪便含水量;可以显著降低结肠黏膜组织中AQP1 mRNA表达;此外,还可以缩短小鼠止血时间和凝血时间。结论 痔断根片具有显著的抗炎、镇痛作用,润肠通便,增加结肠黏膜组织AQP1表达,促进止血,为痔断根片进一步推广提供了依据。

      Objective To evaluate the anti-inflammatory, analgesic, cathartic, and hemostatic effects of Zhiduangen Pian in the treatment of hemorrhoids, and its effect on the expression of Aquaporin 1 (AQP1) in colonic mucosa. Methods The xylene induced ear swelling model was used to study the anti-inflammatory effect of Zhiduangen Pian. The acetic acid induced pain model was used to evaluate the analgesic effect. The constipation mice model induced by loperamide was used to evaluate the peristalsis effect. The expression of AQP1 mRNA in colonic mucosa of constipation mice was detected by real-time fluorescent quantitative PCR. The hemostasis effect was study via hemostasis and coagulation test. Results Zhiduangen Pian can significantly inhibit the swelling of auricle caused by xylene with dosage dependent. Zhiduangen Pian can reduce the times of painful twisting induced by acetic acid, and resist the pain response. Zhiduangen Pian can reduce the cacation time of the first black stool in constipated mice, and increase the number of black stool particles and the water content of feces within 5 hours. Zhiduangen Pian can induce the expression of AQP1 mRNA in colonic mucosa. Zhiduangen Pian can shorten the hemostasis time and coagulation time in mice. Conclusion Zhiduangen Pian has significant anti-inflammatory, analgesic, and laxative effects, and it can increase the expression of AQP1 in colonic mucosa and promote hemostasis. This study provides a basis for further promotion of Zhiduangen Pian.
